In this section, the detailed day-to-day operations are set out with approximate times required for each operation and the optimal periods required between successive operations. Here the actual methods used to obtain reproducible results are noted, although variations in techniques and apparatus can often be satisfactorily substituted. Where possible, alternative techniques have been indicated. Conclusions. This part includes a discussion of the results the student should expect and leads him to draw the correct conclusions from the experiment. General Notes To avoid repetition, the words 'sterile' and 'sterilization' have been omitted from most of the text. It should be assumed that all solutions and glassware used in each experiment are sterile. Moreover, although for most experiments it is not necessary to observe complete sterility, reasonable precautions should be taken, i.e., bottles and tubes should remain uncapped and the agar surfaces of plates should remain exposed for minimal periods. In addition, an incubation temperature of 37°C should be used throughout, unless otherwise stated. All temperatures are expressed in degrees centigrade, shown without C. Genetic loci are usually shown as 3-letter italic symbols [as recently proposed by Demerec et al. (1966) Genetics, 54, 61]. For example, met indicates a methionine-requiring strain (see Appendix В for abbrevia¬ tions).
